105 293T cells were pelleted and the supernatant (media) was removed. Cell pellets were resuspended in 500 µl of DNAgard or water (non-protected). The cells were then spotted on 96-well microtiter plates for drying in a sterile laminar flow hood. For comparison, 105 cells were spotted on Whatman FTA paper and dried. As a positive control, 105 cells were stored at -80°C in 10% DMSO. Dried samples were stored at room temperature or 50°C in heat-sealed pouches containing desiccant. After 65 days of incubation, the dried DNAgard and non-protected samples were rehydrated in 500 µl 1X PBS buffer. These samples, and the -80°C positive controls, were then processed for DNA isolation using a commercially available column purification technology. (The DNA was eluted from the FTA samples using a published protocol.) The DNA yield and integrity from 5 x 104 293T cells was compared directly on a 0.8% agarose gel.
