Stabilization of DNA at Room Temperature - Convenient archiving.
DNAstable Blood provides innovative technology for dry storage of bood samples at room temperature. The DNAstable matrix forms a thermostable barrier round the sample as it dries. Drying occurs in a laminar flow hood at room temperature or in a ventilated 40°C incubator. Samples can then be stored at ambient temperature for at least 12 years (based on accelerated aging theory).

Human whole blood samples (200 μl) were stored for 31 months under various conditions: storage at –20°C (Control); storage in DNAstable Blood at 25°C or 45°C (equivalent to approx. 12 years of storage at room temperature)1; or storage at 45°C without protection (NP). DNA protected using DNAstable Blood is suitable for use in any downstream applications, e.g. long-range PCR, real time PCR, sequencing, etc.

Figure 3A . Successful long-range PCR after 3 years of storage*. Whole blood samples (200 μl) from the same donor were stored for 9 months at –20°C or in DNAstable Blood Tubes at 25°C or 45°C (equivalent to approx. 3 years of storage at room temperature).* After blood rehydration, genomic DNA was purified using the QIAamp® DNA Blood Mini Kit. DNA eluates (2 μl) were then used in PCR of a 5 kb DNA fragment of the human genome using the QIAGEN® LongRange PCR Kit. The PCR products were visualized on a 0.8% agarose gel. M: 1 kb Plus Ladder.

DNA protected using DNAgard is suitable for use in many downstream applications, e.g. long-range PCR, real time PCR, etc. Figure 4. eluates (2 μl) were then analyzed using the artus® MTHFR LC PCR Kit. ‡ Both samples showed no significant difference in melting curve analysis and indicated a heterozygous variant. *Accelerated aging studies: A temperature increase of 10°C corresponds to a twofold
increase in shelf life. For example, DNA stored at 40°C for 2 years corresponds to an
estimated storage time of 8 years at 20°C.
